چکیده

Sugarcane yellow leaf virus (SCYLV) is an aphid-transmitted for which Melanaphis sacchari the main vector. Almost all sugarcane varieties grown in Florida are susceptible to SCYLV infection. In this study, we investigated prevalence of accessions Sorghum bicolor another natural host virus. Two field experiments, one 2016 with 19 sorghum lines and other 2017 15 lines, were established at Belle Glade, FL. Stalks collected randomly planted ratoon crops tested by tissue-blot immunoassay (TBIA) reverse transcription-polymerase chain reaction (RT-PCR). Over two-year period, 366 423 S. samples positive TBIA but was detected RT-PCR only 12 161 selected subsamples. Full genome sequences SCYLV, mosaic (SCMV) a new marafivirus obtained high-throughput sequencing (HTS) from three samples. HTS data viruses confirmed RT-PCR. The SCMV isolates appeared be strain species. Sugarcane Mosaic Virus-Based Gene Silencing in Nicotiana benthamiana

Background:Potyvirus-based virus-induced gene silencing (VIGS) is used for knocking down the expression of a target gene in numerous plant species. Sugarcane mosaic virus (SCMV) is a monopartite, positive single strand RNA virus. Objectives:pBINTRA6 vector was modified by inserting a gene segment of SCMV in place of Tobacco rattle virus (TRV) genome part 1 (TRV1 or RNA1)...

Eradicating Sugarcane Yellow Leaf Virus in Sugarcane Variety N32 through Tissue Culture

Yellow Leaf Syndrome of sugarcane is caused by Sugarcane Yellow Leaf Virus (SCYLV) and/or Sugarcane Yellows Phytoplasma (SCYP). Using tissue blot immunoassay to determine the presence of SCYLV, leaf samples of sugarcane variety N32 taken from a propagation plot at Mount Edgecombe all tested positive. In this study, two methods of tissue culture were used in an attempt to eradicate SCYLV from N3...

Sugarcane Transcript Profiling Assessed by cDNA-AFLP Analysis during the Interaction with Sugarcane Mosaic Virus

Sugarcane mosaic caused by Sugarcane Mosaic Virus (SCMV) is one of the most important virus diseases of sugarcane. In the present study, changes in the transcription profile obtained by cDNA-AFLP analysis were investigated in two sugarcane varieties contrasting to SCMV resistance, when challenged with a severe virus strain. Healthy plants derived from meristem tip tissue culture were mechanical...
